    We introduce SugarQuant, an integrated mass spectrometry-based pipeline comprising protein aggregation capture (PAC)-based sample preparation, multi-notch MS3 acquisition (Glyco-SPS-MS3) and a data-processing tool (GlycoBinder) that enables confident identification and quantification of intact glycopeptides in complex biological samples. Research Summary

Our group focuses on developing cutting-edge mass spectrometry-based technologies for high-throughput yet comprehensive proteomics and protein post-translational modifications profiling of tumors to unveil oncogenic signal transduction and proteomic regulations. Particularly, we take on the analytical challenges arising from the complexity and heterogeneity of glycosylation and develop methodologies for improved glycoproteomic analysis and proteome-wide functional examination of protein glycosylation. Our established workflow allows us to profile proteome, phosphoproteome, and glycoproteome of tumor materials simultaneously. Furthermore, we integrate such multi-layered proteomics with genomics, transcriptomics, and other system-wide functional screens to identify potential therapeutic targets and to support a better prospect of cancer diagnosis and prognosis. In addition, to fully characterize cancer heterogeneity, we are interested in developing single-cell proteomics methodologies and their integration with various single-cell omics technologies. Taken together, we aim to establish versatile proteomic technologies to enable a multi-dimensional molecular characterization of cancers.
